<i>In vitro</i> plant regeneration from different explants of <i>Cardiospermum halicacabum</i> L.

Abstract



A rapid and efficient protocol was developed for inducing indirect organogenesis using stem and leaf explants of Cardiospermum halicacabum L. Explants were cultured on MS medium supplemented with different concentrations of IAA, NAA and 2, 4-D (1.0 to 2.0 mg/l) combined with 0.5 mg/l BAP for callus induction. Callus development and shoot morphogenesis occurred from both explants with varying degrees on different growth regulator concentrations tested. The shoot buds appeared emerging as green coloured protuberances on the callus. The multiplication and elongation of shoots were found to be best on medium supplemented with 2.0 mg/l BAP. Rooting occurred on a medium containing different concentrations of IAA and IBA. Rooted plants were transferred to pots containing sand, soil and manure in the ratio of 1:1:1. Nearly 90% survival of in vitro plants was recorded.
